چکیده

Trypanosoma (Herpetosoma) lewisi is a trypanosome of the sub-genus Herpetosoma (Stercoraria section), parasite of rats (Rattus rattus and Rattus norvegicus) transmitted by fleas. T. lewisi has a stringent species specificity and cannot grow in other rodents such as mice. Rats are infected principally by oral route, through contamination by flea faeces or ingestion of fleas. Trypanosoma lewisi infections in rat colonies can interfere with research protocols and fleas of wild rats are often the source of such infections. Currently, diagnosis of T. lewisi in rats is performed by microscopic observation of stained blood smears. In the course of a research project at CIRDES, a T. lewisi infection was detected in the rat colony. In this study we evaluated PCR primer sets for their ability to diagnose multiple species of trypanosomes with a single amplification. We show that the use of ITS1 sequence of ribosomal DNA provides an efficient and sensitive assay for detection and identification of T. lewisi infection in rats and recommend the use of this assay for monitoring of T. lewisi infections in rat colonies.

Evaluation of trypanocidal drugs used for human African trypanosomosis against Trypanosoma lewisi

Trypanosomes from animals are potential pathogens for humans. Several human cases infected by Trypanosoma lewisi, a parasite of rats, have been reported. The number of these infections is possibly underestimated. Some infections were self-cured, others required treatment with drugs used in human African trypanosomosis. An in vitro evaluation of these drugs and fexinidazole, a new oral drug cand...

Effect of splenectomy on Trypanosoma lewisi infection in young rats.

The effect of splenectomy on animals infected with Trypanosoma lewisi is unclear, and previous reports are inconclusive or conflictive. We splenectomized rats of different ages after they had been infected with T. lewisi. Female Sprague-Dawley rats, Hemobartonella-free, were assigned to four groups according to weight: 80, 108, 140, and 170 g. Each group had splenectomized, sham-operated, and n...

Alpha 2 macroglobulin activity in rats infected with Typanosoma lewisi and treated with cyclophosphamide and its effect on the malignancy of the disease.

BACKGROUND & OBJECTIVES Trypanosoma lewisi is a common, flagellated parasite of the rat. Our previous study showed that rabbits injected with serum collected from rats infected with Trypanosoma lewisi and treated with cyclophosphamide (CyI) produced high levels of antibodies against a new protein in the CyI rat serum. RESULTS In the present study, this protein was characterised as alpha2 macr...

The occurrence and ultrastructure of Trypanosoma (Herpetosoma) lewisi (Kent, 1880) Laveran and Mesnil, 1901, the parasite of rats (Rattus norvegicus) in Poland.

This study reports the light and electron microscopic examination of Trypanosoma (Herpetosoma) lewisi (Kent, 1880) Laveran and Mesnil, 1901, isolated from rats (Rattus norvegicus) from Poland. Bloodstream trypomastigotes were identified morphometrically from 100 specimens collected from three naturally infected rats Rattus norvegicus. Body length ranged from 15.45-23.64 microm and width from 1....

Complement in experimental Trypanosoma lewisi infection of rats.

The role of complement in host resistance to infection with Trypanosoma lewisi was studied in normal, C4-deficient, and C3-depleted rats. Complement levels were measured in normal rats throughout the course of infection. A drastic reduction of total complement and C4 hemolytic activities occurred, and C3 levels measured immunochemically were decreased. Although total complement and C4 levels we...
